What is Microsatellite Instability?
Microsatellite instability (MSI) is a hypermutable phenotype caused by the loss of DNA mismatch repair activity. MSI is detected in about 15% of all colorectal cancers; 3% are of these are associated with Lynch syndrome and the other 12% are caused by sporadic, acquired hypermethylation of the promoter of the MLH1 gene.

Microsatellite Instability (MSI) screening
MSI screening is used to see if the Lynch syndrome genes are working properly. Microsatellites are regions of repeated DNA that change in length (show instability) when mismatch repair is not working properly. MSI testing looks at the length of certain DNA microsatellites from the tumour sample to see if they have gotten longer or shorter as a measure of instability.

Microsatellite instability (MSI) is caused by defects in DNA mismatch repair proteins (MSH2, MLH1, PMS1, PMS2, MSH6, or MSH3). It manifests as an abnormal (increased or decreased) length of microsatellite repeats. The presence of microsatellite instability is a sign of DNA mismatch repair deficiency that can be inherited.
